Annotations for the interaction | figure legend:f1a comment:"\"To understand the fibrillization process in different mixing ratios of Aβ42 to Aβ40, we first used ThT assay to monitor their fibril formation (Figure 1a). ThT is an amyloid fibril probe that emits fluorescence when bound to cross β-sheets of fibrils and its fluorescence intensity indicates relative fibril quantity. Five Aβ samples were prepared in the Aβ42 to Aβ40 ratios as 10/0 (Aβ42 alone), 9/1, 5/5, 1/9, and 0/10 (Aβ40 alone). The total Aβ concentration was fixed at 50 μM. We found that Aβ42 alone aggregated rapidly without a significant lag phase and reached a plateau after ~10 hr of incubation. By contrast, Aβ40 alone has a lag time of ~17 hr and reached a plateau after ~30 hr. By adding 10% Aβ40 to Aβ42 sample (i.e. 9/1 ratio), no significant change in the lag and elongation phases was observed compared to Aβ42 aloneHowever, with addition of 10% Aβ42 to Aβ40 (i.e. 1/9 ratio), the Aβ40 lag time was significantly extended from ~17 to ~80 hr. Interestingly, equally mixed Aβ42 and Aβ40 showed a distinct profile where the ThT signal slightly increased and stayed steady after ~6 hr at relatively low fluorescence (~14.3% of Aβ42).
