Flag-tagged MCM-BP was co-expressed with MCM2-7 proteins in insect cells. Flag-tagged MCM-BP was purified using anti-Flag-agarose beads, and co-purified MCM2-7 proteins were analyzed (Fig. 5). Purified protein complex containing Flag-MCM-BP was separated by glycerol gradient centrifugation. As reported earlier (20), co-recovery of MCM3-7 was observed. Western-blot analysis where purified MCM2-7 proteins were co-electrophoresed shows that MCM3, MCM5 and MCM7 were mainly recovered with MCM-BP and smaller amounts of MCM4 and MCM6 were also recovered, suggesting that MCM-BP has higher affinity for MCM3, MCM5 and MCM7.
